INTRODUCTION
The N-α-Fmoc-α-aminoisobutyric acid derivative (Aib) is a difficult derivative for coupling in SPPS due to its second methyl group on the α-carbon that interferes with the coupling reaction (Figure 1). However, it can be found in a number of biologically relevant compounds, particularly fungal-derived peptide antibiotics such as alamethicin, so methods to successfully couple the Aib amino acid need to be developed. The use of microwave enhanced synthesis helps to drive difficult reactions like those containing the Aib amino acid to completion.
